You don’t have to remove the cap like you did. Just leave it on while priming otherwise more chances of loosing sterility

you can leave the distal cap on. the fluid will still flow, pushing out the air. it will stop flowing on it's own when the line is primed.

you are missing an important step. invert the check valve before priming so it won't trap air and cause air in line alarms.

The key is to release the roller slowly and not all the way unclamped so the soloution primes through the line slowly which prevents bubbles
