i believe peters parents worked for shield or stark industries due to how intelligent Peter is. and they made the spider that bit Peter on his field trip to stark industries. and hydra was trying to get the files to genetically modified spider so they can make supersoldiers so they sent bucky to go kill them and retrieve the files but the files was hidden or was destroyed.
