No. Thor and the rest of the asguardians speak English because they've been with humans for thousands of years. Thor can understand Groot because he speaks Groots language, as suggested by him "taking an elective" on it.

I had a theory on this when I was a kid. My theory was that the producers of movies wanted the viewers to be able to understand the dialog, so they made everyone speak english for movies released in english countries.
