Brain Training Improvement Requires New Myelin This study investigates the requirement for oligodendrocyte (OL) production in non-motor cognition, specifically spatial working memory in adult mice. Using T-maze and 8-arm radial maze tasks, genetic blockade of OL differentiation significantly impaired training-induced performance improvements (control mice achieved ~82% success vs Myrf-cKO ~67% in T-maze; ~31% perfect trials vs ~16% in RAM). Maze training stimulated robust OLP proliferation (~4-fold increase) and new OL generation in key brain regions like the corpus callosum and mPFC. Crucially, individual performance strongly correlated with OLP proliferation (R2=0.84 CC) and new OLs (R2=0.61 CC). Myelination was enhanced, evidenced by a significant ~25% increase in axonal node/paranode structures (6.1 vs 4.8/mm² CC). No positive correlation was found with bulk neuronal activity (c-Fos).
