racecar engineer here. we mark the tires with an arrow pointing to the valve stem to measure slip. Generally, you will find that the front slips more then rear, due to load on fronts under heavy braking. same can be seen in high power rwd cars on the rear wheels under heavy acceleration. for marking the tire, we don't care about slip, the arrow pointing towards valve stem is easy reference for finding valve stem under pit stop tire pressure monitoring / bleeding during test sessions

Bead seal helps it's like a rubber that goes inside the rim where the tire meets
