🧬 For the first time in history, scientists are testing whether aged human cells can become younger again. Every cell in your body carries the same DNA throughout your life. What changes as you age is the epigenome the layer of chemical instructions that tells your genes when to turn on or off. Over time, these instructions drift, and cells gradually lose many of their youthful functions. This breakthrough builds on Nobel Prize-winning research by Shinya Yamanaka, who discovered that four special factors can completely reset mature cells into stem cells. While revolutionary, that approach can erase a cell’s identity, making it unsuitable for many therapeutic applications. A new investigational therapy called ER-100 takes a different path. Instead of using all four Yamanaka factors, it uses only OCT4, SOX2, and KLF4, leaving out the fourth, higher-risk factor. The goal is to partially reset the cell’s biological clock while preserving its original function. In animal studies, this approach restored aging retinal cells and improved vision, building on earlier research from David Sinclair and colleagues published in Nature (2020). On January 28, 2026, the U.S. FDA cleared ER-100 to begin its first human clinical trial. The therapy will be injected into the eye to evaluate its safety and potential effectiveness for treating optic neuropathies. This is the first partial cellular reprogramming therapy to enter human testing. While the results in animals have been promising, it is important to remember that this is an early Phase 1 clinical trial, designed primarily to assess safety. Whether it will work in humans remains unknown.
