We used that technique a long long time ago when we didn’t have access to scanners . You can also get someone to rev it when you pull the connector if the RPM doesn’t drop that cylinder is dead

that trick doesn't work with all vehicles because the ecm can supplement performance. I tried this on a 2010 Honda xt and it did not worsen no matter which cylinder, but the OBD2 accurately states it is po302. the ecm adjusted quickly. and I know it is true misfire because of the fuel trim which strongly suggest injector. so I swapped 2 with 5 and misfire didn't move, so I installed a used injector and adjusted the valves for better clearance.

this is not always the case especially if it's a timing issue that or maf or similar these things will create random miss fires that jump like for example a bmw , oxy and vacuum leaks or low oil pressure causes ignition timing changes that cause eradic misfiring
