@dr.supermom CRISPR technology is a simple yet powerful tool for editing genomes. It allows researchers to study gene function. In the lab, in order to study gene function, we knock-out (delete) the gene of interest using CRISPR/Cas9 technology. We get the lentiCRISPRv2 plasmids containing guide RNA (gRNA) sequences pre-validated by the Broad Institutes. So why we get the lentiviral gRNA expression vector?Because it’s a highly efficient viral vehicle for permanent delivery of target site-specific gRNA sequences into a variety of mammalian cells. Once the gRNA constructs are received in the lab, we perform the following techniques: 1- Plasmid TransformationIt is the process by which foreign DNA is introduced into a cell. Transformation of bacteria with plasmids is important for both storing and replicating plasmids.2- Endotoxin- free plasmid DNA purification. To ensure high yields of high-quality DNA free of contaminants and inhibitors.3- Co- Transfection of Plasmid DNA into HEK 293 Cells Using Lipofectamine and generation of lentiviral particles.4- Lentiviral transductionCells are typically infected in the presence of polybrene, a polycationic chain that neutralizes the charge repulsion between the virus and cell target surface and helps viral integration into the cell. It requires a lot of time, patience and effort especially if you are having many gRNA constructs. It’s worth the effort once you get good results 👍🏻Let me know which technique interests you the most? 😉 The video by @dr.supermom Thank you so much @dr.supermom For this amazing content 📝🙂 . . #biomedicalscience #biologylab #researchlife #research #Science
