hEDS has been classified as a connective tissue disorder, but patients keep reporting immune symptoms (allergies, mast cell issues, autoimmune diagnoses, chronic inflammation) that the connective tissue model doesn’t fully explain. A 2025 study by Griggs et al. in ImmunoHorizons ran an unbiased serum proteomics screen on 29 hEDS patients vs 29 matched controls. They found 35 proteins that were significantly different between the two groups. 43% were part of the complement cascade (a key inflammatory pathway), and 80% were linked to immune, coagulation, or inflammatory pathways. Almost none of the differential proteins were collagen-related. Cytokine profiling confirmed an immune-dysregulation signature. This is single-lab early-stage research that needs replication in bigger and more sex-balanced groups. The study didn’t comprehensively measure all collagen-related proteins, so this finding adds to the picture of hEDS rather than replacing the connective tissue piece. Not a clinical diagnostic.
