Basically fire refers to the fact that Heizou lost his family in a village fire, seeing it with his own eyes, which made him a cold and distant hunter, killing vampires. Paper cranes refer to a Japanese tradition. Folding 1,000 paper cranes grants one wish, or simply means healing and peace. But because Heizou has lost practically everything and his only job is to kill, so he has blood on his hands, it's hard for him to move forward. Blood on paper cranes means that he’s not healing and can’t find peace, so after folding 1,000 paper cranes and soaking them in blood with his own bloody hands, he will never obtain a wish.
