The comic's author gives Dream a human appearance—two arms, a face, hair, clothes, a voice, emotions—because we need to be able to relate to him. But from time to time, the art reminds us that he is not a man. And the eyes are probably the most effective mechanism for doing so. Dream might be sitting and conversing with Hob, he might be angry with Desire, or he might be talking calmly with Death, and then we see those eyes and remember that behind that human appearance lies a much older entity. It's a technique similar to the one the author uses with his language, his posture, and his behavior: humanizing him without letting us forget that he is something cosmic.
