Their relationship hurts my fucking soul. As heartless as he was, Zen really did love his brother but in a twisted and really messed up way. He loved him in the sense that Yoichi was a possession- and one of his first possessions at that. It’s why he wanted so badly to have him returned to him- in his head he “owned” Yoichi- he couldn’t fathom the idea of Yoichi being taken away from him. He loathed this idea so much that when he saw him escaping with Kudo, he killed him for running away from him and then held extreme resentment for Kudo because he genuinely thinks it’s Kudo’s fault that Yoichi got the idea to run away in the first place- so in his head, he didn’t kill Yoichi— Kudo did.

a bit unrelated but I wish the show/manga really emphasised how inhuman afo is. he's basically a robot in a skin suit. for the first 12+ yrs of his life all he did was follow his basic desires, kill steal eat repeat. he literally only had the desire to rule the world because he read a comic book. he has like 0 real emotions and doesn't care at all about anyone except yoichi. this might be a stretch but i always thought that him hunting for yoichi throughout the years could be some sort of metaphor for him chasing his only link to humanity. it's obvious he doesn't really care that much about becoming a demon lord considering he put it on hold for decades just to get yoichi back. it would also explain how much he seems to hate the users of ofa as they are basically obstacles in his way to becoming somewhat human again. it's also a good explanation on why he killed them all so brutally. Another point that adds onto this is how when he realised yoichi was gone he didn't seem to have that much fight in him anymore and then when he heard yoichi's voice he basically begged for him to come back.
