🔬✨ The giant chromosomes of chironomid larvae are a fascinating sight under the microscope ! In the larvae of certain dipteran insects such as Drosophila and chironomids, some tissues (salivary glands, intestine, Malpighian tubules) have cells with giant chromosomes called polytene chromosomes. These special chromosomes were first described in 1881 by French biologist Edouard Balbiani (1822-1899) while he was studying microscopic preparations of chironomid larval tissue stained with methyl green.   Why are they so large? 🤔 Giant chromosomes can reach nearly half a millimeter in length and some 20 µm in thickness, making them easily visible under an optical microscope. Their considerable size is due to the phenomenon of polyteny: instead of dividing normally, the DNA replicates several hundred times without cell division. The chromatids elongate and remain perfectly aligned, making the fine structure of the chromosome much more visible. The result is a much larger chromosome, revealing light and dark bands. Each giant chromosome has a characteristic alternation of transverse bands. The organization of the bands is genetically determined, allowing correlations between the location of the bands and the location of the genes.   These giant chromosomes are used in research to study gene organization and activity.
