Malaria Smear A malaria smear is a microscopic examination of blood to detect Plasmodium parasites in red blood cells. The blood is stained, commonly with Giemsa stain, and examined under a microscope. Types of malaria smear 1. Thick blood film * A larger drop of blood is spread thickly on the slide. * RBCs are lysed during staining. * It is more sensitive for detecting parasites, especially when parasite numbers are low. * Mainly used to determine whether malaria parasites are present. 2. Thin blood film * A small drop of blood is spread into a thin layer. * RBCs remain intact. * It is mainly used for identifying the Plasmodium species and studying parasite morphology. * It can also help estimate parasitemia. Common Plasmodium species * Plasmodium falciparum * Plasmodium vivax * Plasmodium malariae * Plasmodium ovale * Plasmodium knowlesi Basic procedure 1. Collect peripheral blood, commonly in an EDTA tube. 2. Prepare thick and thin blood films on clean glass slides. 3. Allow the films to dry. 4. Stain with Giemsa stain. 5. Examine microscopically, typically using the 100× oil-immersion objective. 6. Look for characteristic parasite forms inside or associated with RBCs. 7. If positive, identify the species and, where required, estimate parasite density. Parasite stages seen Depending on the species and stage, you may see: * Ring forms * Trophozoites * Schizonts * Gametocytes Important: P. falciparum commonly shows delicate ring forms and characteristic crescent/banana-shaped gametocytes, while P. vivax typically infects enlarged RBCs and may show Schüffner’s dots. #malaria #malarias smear #plasmodium #bloodsmear #matiullahdawlatzai
